Principle of the Assay::The Human Visfatin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Visfatin in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human Visfatin coated on a 96-well plate. Standards and samples are pipetted into the wells and Visfatin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Visfatin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Visfatin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Pre-B cell-enhancing factor (PBEF), also designated nicotinamide phosphoribosyltransferase (Nampt) or visfatin, belongs to the NAPRTase family of proteins. PBEF may be involved in enhancing the effect of IL-7 and SCF on the formation of early B-lineage precursor colonies. It is involved in the catalysis of nicotinamide with 5-phosphoribosyl-1-pyrophosphate, yielding nicotinamide mononucleotide, which is important in NAD biosynthesis. This is a rate limiting step in the NAD biosynthesis pathway. Highly enriched in the visceral fat of both human and mice, PBEF expression levels in plasma increase during the development of obesity. PBEF is a cytoplasmic protein expressed primarily in bone marrow, muscle and liver tissue, but it can also be detected in placenta, lung, kidney and heart tissue
